This result suggests that the high expression of miR-71 during L1 diapause is induced or maintained by other signaling pathways. We asked whether the expression of miR-71 was regulated by DAF-16, which is required during L1 diapause for long-term survival (2). It is possible that other miRNAs, including those in the let-7 family, control developmental timing in other tissues during the recovery phase after L1 starvation. We speculate that the expression of heterochronic genes controlling the L2/L3 programs, including that of hbl-1 and lin-42, are increased during L1 diapause to arrest the developmental progression, and miR-71 is probably required to suppress these “excess” signals during the recovery phase (Fig. S5). As pointed out above, multiple miRNAs in addition to miR-71 and the let-7 family miRNAs have roles in L1 diapause, and they may regulate the expression of many diverse targets that may include, but are not limited to, factors involved in UNC-31–InsR-signaling activities. Although the complete removal of miRNA functions causes embryonic lethality or infertility in worms, a partial disruption of overall miRNA functions by mutating either ain-1 or ain-2 provides an effective way to investigate miRNA functions (16, 17).

Unlike dauer diapause, L1 diapause is not accompanied by life cycle changes and has not been shown to require certain signaling pathways that control the formation of dauer diapause such as TGF-β signaling (daf-1, daf-7) and nuclear hormone receptor (daf-12) (2, 3). When late, first larval stage (L1) worms sense unfavorable conditions, they enter an alternative and long-lived larval stage called dauer larvae (or dauer diapause). The nematode Caenorhabditis elegans responds to starvation by entering developmental arrest at multiple stages of its life cycle (1).

Games without play are contests or performances, or both – but not true games. Conversely, having abundant experience of respect and gratitude for fellow players can help us develop habits of moderation, perspective, self-control, and empathy when it comes to non-game frustrations. Rather, we should be – you guessed it – intelligently curating and sharing the more pro-social alternatives, and giving people context and vocabulary for their engagement with culture. Not by running “don’t play Monopoly” campaigns, any more than we run “don’t read mass-produced paint-by-numbers genre pulp that robotically replicates toxic social norms” or “don’t watch reality exploitation TV” campaigns. Our job is to intelligently curate and share the worlds of culture and information in service to our communities, helping people find context and vocabulary for their engagement with the wider world. And as we’ve discussed above, simply offering the kind of safe, supported opportunities to play that we’d want to offer anyway is enough to create that space for healing.
Playing The Tape Forward: A Foundation For My Sobriety
We don’t have to give people everything they ask for; we just have to visibly do our best, care about the result, and be reasonable and respectful about it. I’ve already talked about the links between play and health as part of my Talking Points series – twice, in fact, given that resilience is a key element of mental health. We were rightly concerned that the containment lockdowns would heighten the risk to individuals in abusive domestic situation; this is a clear risk factor. Its own obnoxiousness driving people away from the entire medium only exacerbates this grip; people get a taste and never make it to the good stuff.
We identified a conserved “pause and play” mechanism of cell cycle during environmental stress and recovery. On 31 October, the band announced the 2025–2026 Tanzneid World Tour and released the song «Tanzneid» with a music video featuring Zummo on drums for the first time. In March 2022, they announced that they had changed their name from Eskimo Callboy to Electric Callboy after learning that «Eskimo» can be seen as an outdated term and slur for the Inuit and Yupik people.
It helps maintain a balance where the family supports the recovering addict without enabling their addiction. Individual GFP reporter constructs for candidate genes (4 ng/μL) and the mCherry internal control plasmid (4 ng/μL) were mixed with unc-119 rescuing plasmid (20 ng/μL) and pBluescript KS+ (72 ng/μL) and coinjected into unc-119(ed3) and mir-71(n4115); unc-119(ed3) worms following standard protocols (32). To compare the survival rates between strains, we simulated the survival rate of each genotype to 100 arbitrary “individual worms” and performed the log-rank test in Graphpad Prism 4.
We further examined worms recovering from 4 d of L1 starvation and found that around 90% of the mir-71(lf) mutants displayed retarded vulval precursor cell (VPC) division, compared with less than 5% in wild type (Fig. 4A). In contrast, the nuclear-localized GFP expression under the control of the 3′UTR of age-1(Fig. 3 C and D) or unc-31 (Fig. 3 E and F) was strongly repressed in the control worms, but prominently derepressed in mir-71(lf) mutant worms. If the 3′UTR of age-1 or unc-31 is repressed by miR-71, the GFP expression will be repressed in tissues where miR-71 is expressed in wild-type worms, but derepressed in the same tissues of mir-71(lf) worms. Among short-lived miRNA mutants, a mir-71 deletion mutant, mir-71(n4115) (referred to as mir-71(lf) hereafter), displayed a severe reduction in L1 starvation survival rate (Table S1 and Fig. 2A).
Given my views on play, you won’t be surprised to learn that I asked the presenters about whether play experiences could qualify as the kinds of responsive, positive, engaged, and empowering interactions they described. I was attending professional development aimed at helping library staff support families from refugee backgrounds, and we were discussing the role that libraries and childcare staff (mostly the latter, based on the mix of the group) can play in helping often highly traumatised communities. A small increase in risk of harm, replicated many times on a massive scale, means harm is certainly occurring. As COVID-19 lockdowns unfolded around the world, and sales of games, puzzles, toys, and other playful artforms spiked, a friend asked me if I felt a sense of validation. And worst of all, a combination of the two, where other players essentially become supporting cast for a scripted victory that is neither earned, nor fun, nor remotely playful – a psychodrama rather than a game, toying with their victims rather than playing with them. Controlling bullies who insist everyone play their way, forcing people to make deals they don’t want and to keep playing even when they aren’t having fun.
The coordinated entrance into developmental arrest, long-term survival, and the reinitiation of development upon food availability are important biological processes to investigate. Different organisms have developed versatile growth arrest strategies to overcome starvation-induced metabolic and developmental problems. Food deprivation is a life-threatening challenge that animals frequently face as individuals and as species. Numerous animal species across multiple phyla enter developmental arrest for long-term survival in unfavorable environments and resume development upon stress removal.
